Should there be connectivity between the two wires to a speaker
If there should be it can be checked with a voltmeter ?

Positive and negative to each speaker, no connection between them, you should be able to continuity test from one end of the wire to the other end of the wire.

Some of the speaker wires may be connected together in some sense to have multiple rooms receiving them same signal.
